English. In vitro germplasm conservation is focused on endangered species of medicinal crops. These kinds of crops are usually conserved in collection garden, with high risk of losing by disease infection or bad condition of climate. Ruta angustifolia is classified as endangered species. In vitro conservation has been conducted by maintaining a small part of plant on MS media applied with growth retardant (ABA and Manitol) or by maintaining a growing culture with repeated subculture. Conservation in culture of ABA 4 mg/l and manitol 1000 mg/l gave viability regeneration for six months. Conservation on the growing media of 3/4 MS + BA 0.1 mg/l with subculture after three months gave constant viability after four years of subculture
