[Isolation of proteoglycan compound for toxic Ganoderma lucidum mycelium]

Ganoderma lucidum is a medicinal mushroom which has been used as a health food and need to search its safety through active substances properties. This research is a first stage research which has an objective to produce a safety health food or lead compound. In this paper will be reported: the result of medium selection from 4 isolates of G. lucidum partial purification of glycan extract and the activity of fraction toward Raji Cell line (ATCCCCI-86) and leukemia cell line (L-1210). Isolates were cultured in three liquid media namely : mixed organic-inorganic, malt extract and potato dextrose broth medium. Metabolites was separated from fungi mycelia after five weeks cultured periods. The metabolistes was precipitated using ethanol, centrifuged and determined as a crude glycan. This extract further purify by filtration gel technique using Sephadex G-100. The fraction were collected and grouping based on its similarity in this Rf value. The spot identified using UV light at 256 and 354 nm and specific reagent anyline-hydrogen phatalate mixture. The research result showed that mixed organic-inorganic medium was the best medium for producing metabolites and also glycan crude extract. The highest metabolites and glycan crude extract production found from PSB-5 isolate yield: 9.9932 g/l and 44.4736 g/l respectively. The partial purification using gel, filtration gave 2 fraction which showed the glycan properties (Rf 0.34-0.35 and 0.25). Cytotoxic test using Raji Cell line with 0.15 percent concentration of fraction showed that the fraction with Rf 0.35 from isolates PSB-5 gave the highest activity. This fraction could killed 82.35 percent of cell population at concentration of 14 mg/ml. The other fraction (Rf0.25) from this isolate also gave the highest activity toward leukemia cell line (L-1210) which could killed 80.33 percent of cell population. Based on this result, the next stage research will be focus on the characterization and toxicity of the fraction
