Immunoadsorbent technique for Clostridium perfringens antitoxin purification

English. An immunoadsorbent technique was used for purification of specific antitoxin of Clostridium perfringens. It is based on the insolubilisation of the antigen followed by absorption of the specific antibodies. The red blood cells treated with glutaraldehyde and subsequently with toxins formed sufficient absorptive capacity to be used in purification of the antibody content of the heterologous sera. Treated red blood cells coated with alpha toxin from Cl. perfringens type A were used to absorb anti alpha toxin from B, type C and type D. Anti alpha, anti beta anti epsilon toxin in the sera were measured before and after the absorption process using passive haemaglutination test in the microtitre system.

Teknik ini pada dasarnya adalah insolubilisasi dari antigen yang diikuti dengan absorpsi dari antibodi yang spesifik. Sel-sel darah merah yang telah mendapat perlakuan dengan glutaraldehida dan kemudian toksin, membentuk kemampuan absorpsi yang dapat digunakan dalam pemurnian antibodi dari serum-serum hetorolog. Sel-sel darah merah yang telah mendapat perlakuan dengan glutaraldehida dan toksin alfa Clostridium perfringens tipe A kemudian digunakan untuk mengabsorbsi antitoksin alfa dari anti tipe B, anti tipe C dan anti tipe D. Pada waktu sebelum dan sesudah proses pemurnian, antitoksin-antitoksin alfa, beta dan epsilon dalam serum diukur dengan menggunakan uji hemaglutinasi pasif dalam sistem mikrotiter.
