Mutagenicity of the diethylphosphoric ester of dicyclopropyloketoxime, evaluated by the micronucleus test, on rats

The action of the new organophosporus (OP) compound diethylphosphoric ester of dicyclopropyloketoxime upon cell division of bone marrow (cytogenetic action) was studied and the possible mutagenic action was evaluated by the micronucleus test. The OP compound was administered to male and female Wistar rats by a single oral intubation, at doses of 1.0, 2.5 and 5.0 mg/kg body weight with corn oils as the carrier. The positive controls received a single oral dose, of 20 mg/kg body weight of the mutagenic compound cyclophosphamide (CP). The control animals received only the carrier. The number of micronucleated polychromatic erythrocytes (MN-PCE), the number of micronucleated orthochromatic erythrocytes (MN-NCE) and the number of orthochromatic erythrocytes (NCE) per 1000 polychromatic erythrocytes (PCE) per experimental animal were recorded. The incidence of MN-PCE and the rate of erythrocyte production (% PCE per PCE+NCE) were not affected by the administration of the OP compound, except in the case of female rats at the dose of 5 mg/kg b.w., where a decrease in the above rate was observed. The CP administration increased the incidence of micronucleated PCE and drastically decreased the rate of erythrocyte production. The results indicate the lack of mutagenic action of the OP compound under the experimental conditions of this study.
