Effects of bap concentration on in vitro shoot multiplication and callus induction of Jatropha curcas

The present study optimized the in vitro callus induction from petioles and shoot multiplication from shoot tips in Jatropha curcas. Both explants were inoculated onto MS medium supplemented with 0.5 mg L-1 indole acetic acid (IAA) in combination with different concentrations of 6-benzyloamonipurine (BAP): 0, 0.1, 0.5, 1.0, 1.5 and 2.0 mg L-1. The percentage of callus for-mation in the petioles and shoots proliferated occurred according to the media tested. The highest cell proliferation was obtained from explants cultured in the presence of 1.0 mg L-1 BAP + 0.5 mg L-1 IAA. Out of five media for the shoot multiplication experiment, media with no presence of BAP have no sign of new shoots being produced even though the explants remain green and fresh after eight weeks of inoculation with low percent of petioles forming callus. The highest values of length and number of shoots produced were obtained in the media supplemented with 1.0 mg L-1 BAP + 0.5 mg L-1 IAA.
