Polyploidy induction of the Dendrobium hybrid 'Miss Singapore' through tissue culture

This study was carried out to determine the optimal concentrations of colchicine and incubation time of polyploidy induction in the Dendrobium hybrid 'Miss Singapore' through the tissue culture. Dendrobium 'Miss Singapore' protocorms were treated with colchicine at the concentration of 0, 0.01, 0.05 and 0.1% amended in the MS medium for 1, 2, 3 and 4 days at 25±2°C under dark condition. The results showed that the percentage of surviving plantlets decreased when the colchicine concentration and incubation time increased. The optimal treatment was a colchicine concentration of 0.01% and incubation time of 2 days. Only tetraploids were obtained under such condition. The chromosome number was 2n=4x=76 as compared to the diploid control plantlets 2n=2x=38.
