An analysis of the dietary fiber content of a standard wheat bran

A new procedure for the qualitative and quantitative analysis of the dietary fiber content of a standardized, food grade wheat bran used fractionation and gas-liquid chromatography techniques. The procedure involved fractionation of the freeze-dried, ground wheat bran, extraction with cold and hot water, enzyme treatment of water-insoluble residue, extraction and hydrolysis of hemicellulose and cellulose fractions, determinations of Klason lignin, hydrolysis of water-soluble carbohydrate polymers, analysis of neutral and acidic sugars, and determination of response factors. The wheat bran contained 0.79% cold water-soluble neutral polysaccharides, 2.17% total pectic substances, 28.45% hemicellulose, 9.84% cellulose and 2.87% Klason lignin. These values agreed with those estimated by other methods. The new procedure produces a quantitative profile of the major components of dietary fiber, so that comparison of the fiber content of foodstuffs is possible.
