Characterization by 1H NMR spectroscopy of enzymically derived oligosaccharides from alkali-extractable wheat-flour arabinoxylan

Oligosaccharides obtained from alkali-extractable wheat-flour arabinoxylans by digestion with endo- (1 leads to 4)-beta-D-xylanase from Aspergillus awamori were fractionated by size-exclusion chromatography on Bio-Gel P-2 followed by high-performance anion-exchange chromatography, and subjected to monosaccharide analysis and 1H NMR spectroscopy. The results revealed (1 leads to 4)-linked beta-D-xylopyrano-oligosaccharides partly 3- and/or 2,3-substituted with single alpha-L-arabinofuranosyl groups. The structures of 12 such arabinoxylan oligosaccharides were established.
