Effect of ethanol on the disposition of paraquat

Rabbits that received paraquat (200 mg/kg) and ethanol (1.0-3.8 g/kg) by gavage resulted in shorter median survival times and higher mortality rates than those dosed with paraquat alone. Plasma paraquat levels in rabbits dosed with ethanol before, after and concurrently with ingestion of paraquat were significantly higher when compared with animals without ethanol intake. Furthermore, substantial differences in paraquat kinetics, higher AUC, lower total clearance (CL/F) and volumn of distribution (V/F) were seen in the ethanol-paraquat group. The effect of ethanol on acute paraquat toxicity is, at least in part, associated with the increase in paraquat absorption and/or the decrease in paraquat excretion.
