Cholesterol versus other sterols: How do they compare as physiological regulators of cholesterol homeostasis?

There is a growing body of work indicating that maintaining cholesterol homeostasis involves oxysterol‐derivatives and sterol precursors of cholesterol. Their importance relative to cholesterol itself is a contentious area, which is difficult to address experimentally. In this review, the molecular basis of cholesterol homeostasis is briefly described, some of the major putative sterol regulators introduced, and the evidence that they may indeed serve as physiological regulators is discussed, along with caveats of this research. This review will also briefly address some recent literature ascribing physiological roles of non‐cholesterol sterols beyond cholesterol metabolism.
