Determination of uronic acids in soil hydrolysates by quantitative reduction to the aldonic acids and separation of their aldono-1,4-lactone derivatives by high-performance gas chromatography (HPGC) proved to be a very effective method. Identification and quantitation of glucuronic (GlcA) and galacturonic (GalA) acids was possible without prior separation from neutral and amino sugars. The m-hydrox-ydiphenyl colorimetric procedure gave a fast and reliable estimate of total uronic acids in soil hydrolysates. A method based on esterification of uronic acid residues followed by reduction to the corresponding hexoses was found to be unsuitable for measuring uronic acids in soils.
