Analysis of Genetic Diversity of Actinidia chrysantha Conserved in the National Guangdong Nanling Natu ral Reserve by ISSR Markers

Actinidia chrysantha has not been exploited as a fruit crop and the scientific research at DNA level is also less documented. The national Guangdong Nanling Natural Reserve is one of places where A. chrysantha germplasm is well present. In order to preserve and further develop these precious and endangered kiwifruit resources, the genetic diversity of 16 individuals from four populations of A. chrysantha collected in the Guangdong Nanling national reserve was analyzed by ISSR markers, and the UPGMA clustering was carried out on the amplificated bands of ISSR markers. Nine primers could amplify 196 DNA loci, of which 195 were polymorphic, representing 99.5% of all amplified loci. The average number of polymorphic loci for each primer was 21.8, the highest number being obtained with primer UBC812 that gave 30 loci in total. Considering in the first approximation the bands in the gel as unique loci, the polymorphism information content (PIC) was 1.99, 1.39, 0.25, 0.41, 0.93, respectively for each population. According to the UPGMA clustering most of the A. chrysantha plants in the same populations clustered in the same group, suggesting that the common geographic origin had the greatest effect in explaining the genetic diversity of the sampled accessions. The results showed also that A. chrysantha in Guangdong Nanling Natural Reserve has a high genetic diversity.
