Genetic diversity of Verticillium dahliae isolates from olive trees in Algeria [Olea europaea L.]

English. Verticillium wilt of olive trees (Olea europaea L.), a wilt caused by the soil-borne fungus Verticillium dahliae (Kleb), is one of the most serious diseases in Algerian olive groves. To assess the pathogenic and genetic diversity of olive-infecting V. dahliae populations in Algeria, orchards from the two main olive-producing regions (North-Western Algeria and Kabylia) were sampled and 27 V. dahliae isolates were recovered. For purposes of comparison, V. dahliae strains from France and Syria were added to the analysis. By means of PCR primers that specifically discriminate between defoliating (D) and non-defoliating (ND) V. dahliae pathotypes, it was shown that all V. dahliae isolates belonged to the ND pathotype. The amount of genetic variation between the 43 isolates was assessed by random amplification of polymorphic DNA (RAPD). A total of 16 RAPD haplotypes were found on the basis of the presence or absence of 25 polymorphic DNA fragments. Genotypic diversity between the 27 Algerian isolates was low, with two RAPD haplotypes accounting for 70% of all isolates. Genotypic diversity was however greater among isolates from Kabylia than within isolates from North-Western Algeria. Cluster analysis showed that most of the Algerian V. dahliae isolates grouped together with the French and Syrian isolates. On the basis of their ability to form heterokaryons with each other, a subset of 25 olive-pathogenic isolates was grouped into a single vegetative compatibility group (VCG). These results suggest that the olive-infecting V. dahliae populations in Algeria show limited diversity and that caution should be taken to prevent introduction of the D pathotype

[La verticilliosi dell'olivo (Olea europaea L.), causata dal fungo trasmesso dal terreno Verticillium dahliae (Kleb), è una delle malattie più gravi negli oliveti algerini. Per accertare la diversità genetica e di patogenicità delle popolazioni di V. dahliae infettanti l'olivo in Algeria, sono stati effettuati campionamenti negli oliveti delle due regioni più importanti per l'olivicoltura (Algeria Nord-Occidentale e Kabylia) e sono stati raccolti 27 isolati di V. dahliae. A scopo di confronto, sono stati aggiunti all'analisi ceppi di V. dahliae francesi e siriani. Per mezzo di primer PCR in grado di discriminare fra patotipi di V. dahliae defolianti (D) e non defolianti (ND), è stato dimostrato che tutti gli isolati di V. dahliae appartenevano al patotipi ND. Il livello di variazione genetica fra i 43 isolati è stato accertato mediante amplificazione a caso del DNA polimorfico (RAPD). Sono stati trovati, in totale, 16 aplotipi RAPD in base alla presenza o assenza di 25 frammenti di DNA polimorfico. La diversità genotipica fra i 27 isolati algerini è risultata bassa, con due aplotipi RAPD che incidevano per il 70% di tutti gli isolati. La diversità genotipica è comunque risultata maggiore nell'ambito degli isolati della Kabylia rispetto a quelli dell'Algeria Nord-Occidentale. L'analisi cluster ha mostrato che la maggior parte degli isolati algerini di V. dahliae si raggruppa assieme agli isolati francesi e siriani. In base alla rispettiva capacità di formare eterocarionti l'uno con l'altro, un sottoinsieme di 25 isolati patogeni per l'olivo è stato raggruppato in un singolo gruppo di compatibilità vegetativa (VCG). Questi risultati fanno concludere che le popolazioni di V. dahliae infettanti l'olivo in Algeria presentino una diversità limitata e che dovrebbero essere prese precauzioni per prevenire l'introduzione del patotipo D.]
