PCV2 detection in 170 submaxillary lymph nodes of regularly slaughtered pigs in Northern Italy [Porcine Circovirus Type 2]

English. One-hundred-seventy submaxillary lymph nodes from regularly slaughtered pigs from 28 different farms of Northern Italy were randomly sampled. The aim was to evaluate the presence of PCV2 in slaughtered pigs, since serological investigations are not helpful to determine the state of the infection at the time of slaughter. Follicular lymphoid hyperplasia was detected in all cases. Seven cases showed a concomitant granulomatous lymphadenitis with necrotic foci. Chronic lesions consistent with Post-weaning Multisystemic Wasting Syndrome (PMWS) were not detected in the lymph nodes. Immunohistochemistry (IHC) employed the PCV2-specific monoclonal antibody, which reacts with the virus capsid antigen. Fifty-seven out of 170 lymph nodes (33.5%) were positive to PCV2. Staining was always at the centre of the lymphoid follicles and the reactivity was detected in the cytoplasm of follicular dendritic cells, macrophages and lymphocytes. In these cases, in situ hybridisation (ISH) protocols were developed, employing a probe which maps the ORF1 (Open Reading Frame 1). It encodes a replication-associated protein (Rep) essential for replication of viral DNA. Out of 57 IHC positive cases, only four were positive to ISH (7%), with rare and specific lymphoid follicular spots. Lymph nodes positive to IHC and negative to ISH were interpreted as cases in which an exposure of PCV2 had occurred, whereas four cases also positive to ISH suggested an active viral infection

Sono stati campionati random 170 linfonodi sottomascellari di suini provenienti dal Nord Italia (28 allevamenti diversi), macellati regolarmente, allo scopo di valutare la presenza di PCV2 in soggetti in cui, stante la diffusione del virus, l'indagine sierologica non fornisce indicazioni sullo stato di infezione al momento della macellazione. Nella totalità dei casi era presente solo iperplasia follicolare e in 7 casi vi era anche una linfadenite granulomatosa con focolai di necrosi istologicamente non compatibili con lesioni croniche da Post-weaning Multisystemic Wasting Syndrome (PMWS). Inizialmente si è utilizzata l'immunoistochimica (IHC) per determinare la presenza della proteina capsidica del Circovirus di tipo 2: sono stati riscontrati 57 linfonodi positivi su 170 (33,5%). La positività era sempre centrofollicolare; le cellule positive, riconosciute in base alla morfologia, erano cellule follicolari dendritiche, macrofagi e linfociti. La positività era di tipo citoplasmatico. Nei casi suddetti è stata applicata la metodica di ibridazione in situ (ISH), utilizzando una sonda per l'ORF1 (Open Reading Frame 1), che individua una proteina rep associata alla replicazione. Dei 57 casi positivi all'IHC, solo quattro casi (7%) presentavano all'ISH rari ma specifici spot centrofollicolari. Sulla base dei risultati ottenuti, i linfonodi positivi all'IHC e negativi all'ISH sono stati interpretati come provenienti da animali in cui si è verificata una esposizione al PCV2, mentre i quattro casi positivi anche all'ISH suggeriscono una infezione virale attiva
