Monoclonal antibody capture fluorometric enzyme-linked immunosorbent assay for detection of equine growth hormone in plasma

English. The enzyme-linked immunosorbent assay (ELISA) developed is based on a monoclonal antibody (mAb) with high affinity to equine GH (eGH) and highly purified pituitary eGH. The mAb was used as solid-phase antibody to capture eGH, followed by biotin labelled polyclonal antibodies and fluorescent detection. The standard curve was linear between 0.05-10 ng/ml. Plasma did not interfere with the ability to measure eGH. The intra- and inter-assay CV ranged between 4.4-15.0% and 14.0-20.0%. The analytical specificity was validated by provocative test using the releasing peptide GHRP-6 in horses. In conclusion, the ELISA is sensitive, reproducible and suitable for measuring GH in horse plasma, and offers an alternative to RIA

[Il metodo immunoenzimatico ELISA sviluppato è basato su un anticorpo monoclonale (mAb) con elevata affinità per il GH equino (eGH) e per un eGH ipofisario altamente purificato. L'mAb è stato utilizzato come anticorpo in fase solida per catturare l'eGH, seguito da anticorpi policlonali marcati con biotina e da rivelazione in fluorescenza. La curva standard è risultata lineare fra 0,05 e 10 ng/ml. Il plasma non interferiva con la capacità di misurazione dell'eGH. Il CV inter- e intra-dosaggio variava fra 4,4-15,0% e fra 14,0-20,0%. La specificità analitica è stata validata mediante un test di stimolazione in vivo con il peptide GHRP-6.
