Evidence of non-transmission of Rice yellow mottle virus through seeds of wild host species [Oryza sativa L.; Burkina Faso; Chad; Mali]

English. Rice yellow mottle virus (RYMV) is a widespread and damaging rice pathogen in Africa. The presence, infectivity and transmissibility of RYMV in seeds of six wild host species were investigated. In serological analyses by enzyme-linked immunosorbent assay (ELISA), each of four RYMV isolates used was detected in individual seeds of wild rice species (Oryza barthii and O. longistaminata) and the standard susceptible rice BG 90-2 (O. sativa). Virus detection in seeds of other host species (Dactyloctenium aegyptium, Eragrostis ciliaris, E. tenella and E. tremula) was unsuccessful in single seeds, but positive reactions were obtained for extracts made from several seeds. RYMV was infectious in freshly-harvested seed extracts whatever the plant species or the virus isolate. However, most infectivity was lost in dried seeds, possibly due to virus inactivation following dehydration of the seeds. Despite the presence of RYMV in dry seeds, no evidence was obtained for transmission of the virus through seeds of rice or wild host species, whatever the plant species-virus isolate combination. RYMV is unlikely to be transmitted through seeds of its host species and, therefore, virus dissemination or epidemics of rice yellow mottle do not originate from infected seeds

Sono state studiate la presenza, l'infettività e la trasmissibilità di RYMV nei semi di sei specie ospiti spontanee. In analisi sierologiche mediante ELISA, ognuno degli isolati di RYMV utilizzati è stato riscontrato in semi individuali di specie selvatiche di riso (Oryza barthii e O. longistaminata) e nel riso suscettibile standard BG 90.2 (O. sativa). La rilevazione del virus in semi di altre specie ospiti (Dactyloctenium aegyptium, Eragrostis ciliaris, E. tenella e E. tremula) non ha dato esito positivo nei semi singoli, ma sono state ottenute reazioni positive per estratti di numerosi semi. RMYV era infettivo negli estratti di seme appena raccolti, indipendentemente dalla specie di pianta o dall'isolato del virus. Tuttavia, la maggior parte dell'infettività veniva perduta nei semi essiccati, possibilmente per l'inattivazione del virus a seguito della disidratazione dei semi. Nonostante la presenza di RYMV nei semi secchi, la trasmissione del virus fra semi di riso o specie ospiti spontanee non è stata provata, in tutte le combinazioni specie-isolato del virus. E' improbabile che RYMV sia trasmesso da semi delle sue specie ospiti e, perciò, la diffusione o l'epidemia di maculatura gialla del riso non si origina da semi infetti.]
