English. Praziquantel (PZQ) is an anthelmintic drug widely employed in veterinary medicine. Several metabolic studies have been performed in both laboratory animals and in humans, but no data are available in ruminants. Because of several differences in isoenzyme P450 dependent system among humans, rodents and ruminants, and of PZQ fast and intensive metabolism, the present study was undertaken to determine the involvement of the cytochrome P450 in the PZQ metabolism and to evaluate the formation of its metabolites in 3 month old lambs. The 3A dependent activities were inhibited after the administration of the PZQ at 60 mg/kg/die for 2 days and the P450 3A resulted to be the primary isoform responsible for the transformation of PZQ in PZQ 11b-OH, which is the main metabolite in lambs

Sono stati effettuati numerosi studi metabolici sia su animali da laboratorio, sia sull'uomo, ma non sono disponibili dati riguardanti i ruminanti. A causa delle numerose differenze nel sistema di isoenzimi P450 dipendenti fra l'uomo, i roditori e i ruminanti, e per il metabolismo rapido e intensivo di PZQ, e' stato intrapreso il presente studio per accertare il coinvolgimento del citocromo P450 nel metabolismo di PZQ e per valutare la formazione di suoi metaboliti in agnelli di tre mesi.
