Differentiation of Drechslera graminea isolates by cultural characteristics and SDS-PAGE [Hordeum vulgare L. - Syria]

Isolates of Drechslera graminea, the causal agent of the barley leaf stripe disease, were obtained from phytopathological samples collected in 1998 from different regions of Syria and analyzed for differences in cultural characteristics and protein profiles as determined by a 10% sodium-dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). Differences in mycelia growth and colour were noticed among isolates collected from different regions. On the other hand, protein polymorphisms (SDS-PAGE) were detected and highly significant differences (P 0.001) for band numbers were obtained among isolates. The protein profiles were highly repeatable, suggesting that the biochemical SDS-PAGE protein markers are suitable to detect variability among D. graminea isolates

[Ceppi isolati di Drechslera graminea, agente causale della striatura fogliare dell'orzo, sono stati ottenuti da campioni fitopatologici raccolti nel 1998 in diverse regioni della Siria e analizzati per le differenze nelle caratteristiche di crescita e per i profili delle proteine con elettroforesi SDS-PAGE con gel di poliacrilamide al 10% di sodio dodecilsolfato. Sono state notate differenze di crescita e di colore tra i diversi ceppi isolati nelle diverse regioni. E' stato anche scoperto un polimorfismo delle proteine e differenze altamente significative tra il numero delle bande. I profili delle proteine sono risultati molto ripetibili, suggerendo che i marcatori chimici delle proteine (SDS-PAGE) sono adatti per determinare la variabilita' tra i ceppi di D. graminea]
