Genetic diversity in selected Lathyrus species revealed by restriction fragment length polymorphism (RFLP) markers [Tunisia]

English. A study of the genetic diversity in Lathyrus by RFLP analysis was performed for 137 individuals representing five species (L. sativus, L. cicera, L. ochrus, L. sylvestris and L. latifolius). Nine populations representing these species from various geographical origins were used. Seven DNA clones selected from a genomic DNA library of Lathyrus and a maize 25S ribosomal DNA probe were used for Southern hybridization. Twenty-two enzyme-probe combinations were studied to estimate the genetic variation within and among the studied populations. A total of 103 polymorphic bands were revealed. Based on RFLP data, genetic distance among and within populations was estimated by calculating the dissimilarity index. Cluster analysis and factorial analysis of correspondences showed a clear genetic separation of the population of L. ochrus from the other populations. In addition, L. sylvestris and L. latifolius populations formed one cluster and L. sativus and L. cicera populations formed another cluster

[E' stato condotto uno studio della diversita' genetica in Lathyrus mediante analisi RFLP su 137 individui rappresentanti cinque specie (L. sativus, L. cicera, L. ochrus, L. sylvestris e L. latifolius). Sono state utilizzate nove popolazioni di diversa provenienza geografica rappresentanti queste specie. Per l'ibridazione sono stati utilizzati sette cloni di DNA selezionati da una collezione di DNA genomico di Lathyrus e una sonda di DNA ribosomiale 25S di mais. Sono state studiate ventidue combinazioni enzima-sonda per stimare la variazione genetica entro e fra le popolazioni studiate. E' stato individuato un totale di 103 bande polimorfiche. In base ai dati RFLP, la distanza genetica fra ed entro le popolazioni e' stata stimata calcolando l'indice di disuguaglianza. L'analisi cluster e l'analisi fattoriale delle corrispondenze hanno dimostrato una chiara separazione genetica della popolazione di L. ochrus dalle altre popolazioni.
