Chromatographic analysis of flavonoids in vegetables and their products

English. A method was developed for the determination of flavonoids present in fresh and processed fruit and vegetable products. The flavonoids were extracted using a mixture of dimethylsulphoxide/ethanol (1/19 v/v) at ambient temperature and analysed by HPLC with reverse phase C18 column. This method was applied to the characterisation of the phenolic component of fresh products such as tomato cultivars and processed products such as orange, apple and pear juices. The data obtained were compared with those reported in literature and with those obtained using official methods. As to tomatoes, it is to be underlined that with this method it was possible to determine rutin both qualitatively and quantitatively. This has never been reported in literature and only after hydrolysis quercetin the corresponding aglycone, was determined. As to juices, the obtained data were compared with those resulting from the use of a standardised method; the comparison was made using the statistical test of difference of means and the two methods gave similar results. The method proposed in this work proved to be rapid and reliable; the analytical results obtained are repeatable and reproducible
Show more [+] Less [-]Italian. E' stato messo a punto un metodo per l'estrazione e la determinazione di flavonoidi presenti in prodotti ortofrutticoli freschi e trasformati. I flavonoidi sono stati estratti con l'utilizzo di una miscela di dimetilsolfossido/etanolo (1/19 v/v) a temperatura ambiente e analizzati mediante cromatografia liquida ad alta prestazione (HPLC) con colonna C18 a fasi inverse. Questo metodo e' stato applicato per la caratterizzazione della componente fenolica di prodotti freschi quali cultivar di pomodoro e di prodotti trasformati quali succhi di arancia, mela e pera. I dati ottenuti sono stati confrontati con quelli riportati in letteratura e con quelli ottenuti con l'utilizzo di metodi stabiliti mediante norme. Per quanto riguarda il pomodoro, si sottolinea che e' stato possibile con questa metodica determinare quali-quantitativamente la rutina. In letteratura cio' non e' stato mai riportato e solo in seguito a idrolisi e' stata determinata la quercetina, l'aglicone corrispondente. Relativamente ai succhi, sono stati confrontati i dati ottenuti con quelli corrispondenti all'utilizzo di un metodo normato; il confronto e' stato effettuato con l'ausilio del test delle ipotesi sull'uguaglianza delle medie e i metodi hanno dato risultati simili. Il metodo proposto in questo lavoro risulta essere rapido e affidabile: i risultati analitici ottenuti sono ripetibili e riproducibili
