Culture conditions for the initiation of embryogenesis from isolated microspores in Chinese cabbage (Brassica campestris L.)

Culture conditions of microspores of Chinese cabbage were investigated. The medium which was composed of the vitamins and half strength macronutrients of Nitsch and Nitsch medium (1967), micronutrients of MS medium (1962), 10% sucrose and several other organic compounds, was used after filter sterilization. Microspore suspension at a density of 0.5 to 1 x 100000/ml was incubated at 33 degrees C for 1 day and then maintained at 25 degrees C in the dark. Microspores isolated from buds 2.0 to 2.5 mm in length and with a petal/anther ratio of 0.62 could be used for the production of embryos.
