Enzymecytochemical localization of 5'-nucleotidase in the molecular layer of rat cerebellar cortex

The localization of 5'-nucleotidase was electron microscopically examined in the molecular layer of rat cerebellar cortex using the improved cytochemical method with cerium as a capture reagent recently introduced by Robinson and Karnovsky. Reaction products demonstrating 5'-nucleotidase activity were present on the outside of the neuropil plasma membrane, and per- and postsynaptic membranes, but not on the glial cell membrane. These findings indicate the existence of 5'-nucleotidase on the neuronal plasma membrane as an ectoenzyme, and are different from its previous cytochemical localization reported with methods using lead as a capture reagent, that is, the enzyme is recognized on the plasma membrane of glial cells but not on every neuronal plasma membrane. Considering significant drawbacks of the lead-based method, the cerium-based method offers more accurate results fro ultracytochemical localization
