Induction of embryogenic callus and plant regeneration by somatic embryogenesis from rice [Oryza sativa] anther

Embryogenic calli were induced from the anther of rice (cv. Taichung 65) cultured on the three kinds of media. The three conditions of the media are as follows, that N6 salt supplemented with 2,4-D (2mg/l), yeast extract (1.36g/l), casein hydrolyzate (300mg/l), sucrose (30g/l) and agar (10g/l) was used as a basal medium and NAA (3mg/l) + kinetin (2mg/l), NAA (3mg/l) + kinetin (3mg/l), NAA (3mg/l) + kinetin (4mg/l) were supplemented, respectively. The calli were compact in appearance and white to pale-yellow in color. All of the calli regenerated on the regeneration medium. Regenerated plantlets were bipolar with plumule and radicle in the opposite sides, which was phenotypically similar to the seed derived plants. Embryo-like structures with coleoptile, root meristem and scutellum were formed on the embryogenic callus surface during regeneration
