The southern blot analysis of mouse TGFalpha gene expressions in normal and neoplastic mammary glands

Mouse transforming growth factor alpha (TGF-alpha) gene expression was determined by the reverse transcriptase-polymerase chain reaction (RT-PCR) method in mammary glands and mammary tumors in four strains of mice, C3H/He, SHN, SLN and GR/A. Both normal pregnant and tumorous mammary tissues expressed mRNA for mouse TGF-alpha. There were obviously different PCR products between pregnant mammary glands and tumors. In tumors, various sizes of PCR products were detected in addition to a predicted size. In southern blot analysis, there was a single hybridized band in all RT-PCR samples of both normal and neoplastic mammary glands. In genomic DNA, there was also single band in EcoRI restriction enzyme digestion. Possibly there is only a single copy of TGF-alpha gene in mice, and multiple mRNA expression in tumors seems to be due to the different gene expression products containing similar sequences to mouse TGF-alpha primers
