Analysis of the genetic diversity of traditional fig-tree (Ficus carica L.) cultivars using DNA markers [Portugal]

Nine traditional fig tree cultivars, included in a field collection mainly constituted by germplasm of Portuguese origin, were analysed by RAPDs and AFLPs. From previous molecular studies it was known that these cultivars share identical patterns for six isozyme systems (LAP, GOT, MDH, IDH, PGI and PGM). RAPD analyses were performed using 37 RAPD promers, which revealed 84 (33,5%) polymorphisms among the 251 markers scored. Eight EcoR I/Mse I primer combinations were used in the AFLP analysis revealing 400 markers of which 172 (43%) were polymorphic. Seven out of the nine fig cultivars were discriminated by both techniques whereas between the remaining two cultivars no polymorphic marker was detected. These last data point to a synonymy between these two cultivars: Urjal and Bacorinha. A high correlation (r=0,86) was found between the similarity matrices based on RAPD and AFLP data, confirming the consistency of the genetic relationships established in this study
