[Morphogenetic response of tobacco tissues after infection of leaf with Agrobacterium tumefaciens, strains bearing either a wild or a disarmed Ti plasmid]

The genetic transformation system of plant cells mediated by the bacteria A. tumefaciens has been intensely studied because of the possibilities to be used to obatain transgenic plants with new genetic information incorporated. Also this system offers advantages for analyzing some aspects of plant-pathogen interaction as it is the influence of hormones produced by bacteria that affects the development of plant tissues. In this research we evaluated the morphogenetic response of tobacco tissues in vitro after infection of leaf discs with A. tumefaciens. Strains bearing either a wild or a disarmed Ti plasmid were used. When the leaf discs were infected by strains with the wild plasmid, a tumoral growth was induced with capacity to growth on a free-hormone medium, but without a capacity to produce shoots. Aditionally, it was demostrated that discs infected by the strain with the disarmed plasmid were capable to produce shoots in a medium with or without hormones.

El sistema de transformacion genetica de celulas vegetales mediado por la bacteria A. tumefaciens ha recibido considerable atencion por las posibilidades que tiene de ser utilizado para la obtencion de plantas transgenicas con nueva informacion genetica incorporada. Este sistema tambien ofrece ventajas para analizar algunos aspectos de la interaccion planta-patogeno, como la influencia de las hormonos producidas por la bacteria y que afectan el desarrollo de los tejidos vegetales. En este trabajo se evaluo la respuesta morfogenetica de los tejidos de tabaco bajo cultivo in vitro despues de haber infectado discos de hoja con una cepa de A. tumefaciens portadora de un plasmido Ti silvestre desarmado. Cuando los discos de hoja se infectaron con la cepa portadora del plasmido silvestre, se indujo un crecimiento tumoral capaz de mantenerse en un medio sin hormonas, pero sin capacidad para producir brotes. Se demostro, ademas, que los discos infectados con la cepa portadora del plasmido desarmado fueron capaces de producir brotes en un medio de cultivo con o sin hormonas.
