Some adverse effects of long term administration of the antiinflammatory ibuprofen in mice [Egypt]

Fifty mice of both sexes weighing 15-25 g were used. Thirty mice were given ibuprofen (50 mg/ kg B.Wt. daily) by a stomach tube for three months. The other mice were kept as controls. Ibuprofen decreased blood serum levels of total proteins and calcium and increased alkaline phosphatase (AP), glutamic oxalacetic transaminase (GOT) and glutamic pyrovic transaminase (GPT). Ibuprofen sharply decreased the erythrocytic count, haemoglobin level and packed cell volume. On the contrary, the leukocytic count was moderately increased in the treated mice. Histopathological examination revealed bronchopneumonia, congestion in the portal veins and liver sinusoids and focal encephalomalacia in the cerebrum and cerebellum of treated mice. It is preferable to use alternative treatment of ibuprofen with similar antiinflammatory analgesic drugs if prolonged treatment of such drugs is indicated.
