Molecular genetic studies on Trichoderma sp. related to medical plants in Sinai

Eight strains of Trichoderina (Trl, Tr2, Tr3, Tr4, Tr5, Tr6, Tr7 and Tr8) belong to five species (Trichoderma sp. T. aureoviride, T atroviride, T viride, and T koningii) were tested against plant pathogenic fungi (Fusarium oxysporum). In vitro test of the previous isolates that the Trl, Tr2, Tr3, Tr5and Tr6 is most efficient strains. All isolates were evaluated for production of chitinases and cellulases. Using colloidal chitin as a substrate, Trichoderma atroviride (Tr2) strain was the most efficient in chitinase activity (2.53 u/ml/min), while Trichoderma atroviride (Tr6) strain was the most efficient in cellulase (3.9 u/ml/min) using carboxy methyl cellulase (CMC) in mineral liquid medium. The selected strains have been mutagenized to obtain variants capable to producing high levels of chitinases or cellulases enzymes by using EMS (Ethyl methane sulfonate) 300 ug/ml in two steps of mutagenized. Ten morphogical mutants from parental strains Tr2 and Tr6 were isolated on mineral medium supplemented with colloidal chitin and CMC. The best mutant for chitinase overproduction is (Tr2msl0.10) mutant strain. It produced (5.9 u/ml/min) which equal 236%. On the other hand, the best selected mutant strain for cellulase was (Tr6us2.1) overproduction. It produced (5.3 u/ml/min) which equal 135%. This improvement in extracellular enzyme production by the mutant T atroviride and production protoplast fusion suggests that it is a suitable strain to be used in biological control.
