Identification of diethylstilbestrol, dienestrol and hexestrol in mixed feed by thin-layer chromatography and spectrophotometry U.V. light

A procedure is described for the detection of diethylstilbestrol (DES), dienestrol (DIEN) and hexestrol (HEX) in mixed feeds. The separation is made by Thin-Layer Chromatography and the identification by comparation of Rf with reference standards. The spots are visualized by exposing the plate to U.V. light of 254 nm wavelength and a fluorescence reaction was induced by spraying with 5 p. 100 sulfuric in acetic anhydride. The plate was viewed under U.V. light (366 nm) and then incubated at 95 degrees C during 12 min. The fluorescence was observed under U.V. light of 366 nm wavelength. The spots are also coloured by spraying with 20 p. 100 antimony pentachloride in carbon disulfide. The spots separated by T.L.C., are eluted with ethanol and this solution is spectrophotometrically comparated in the U.V. spectrum area witn ethanolic solution of reference standard
