DPPH radical-scavenging activity of pasteurized cow milk

Pasteurized milk samples of most representative producers in Serbia were collected from local shops. They had different fat content, and some of them were enriched with vitamins (A+D3). The reduction of DPPH (60 microM) in methanol was followed by monitoring a decrease of its absorbance at 515 nm after 45 min. Significant negative correlation (r=-0.85) was obtained between (DPPH) radical-scavenging activity and fat content, which means that antioxidative capacity rises with fat content. In milk sample with 2.8% fat, EC50 is 3.89 ml milk/mg DPPH. Higher antioxidant activity was gained in milk with the addition of vitamins (A+D3). The EC50 value for ascorbic acid was 37.5 microM. DPPH method was very useful in research of the total antioxidative capacity of milk without distinguishing the contributions from individual compounds.
