Detection of genes encoding the wheat technological quality by DNA markers

The aim of our work was to analyse a set of 33 new wheat genotypes and wheat cultivars for the presence of DNA marker which is linked with gene encoding HMW subunit 1Dx5 associated with subunit 1Dy10. The allelic pair 1Dx5+1Dy10 has positive effect on wheat technological quality. Analysis were performed based on D Ovidio et al. (1994) by PCR using two specific primers and obtained DNA fragments were detected in agarose gels stained by ethidium bromide and visualised by UV light. A clear PCR product of 450 bp, representing 1Dx5 of the Glu-D1 alleles, was identified in 26 new wheat genotypes and cultivars. PCR analysis of 1Dx5 were compared with protein analysis done by SDS-PAGE. PCR analysis for gene controlling bread-making quality confirmed that primers used in the present study are specific for the Glu-D1d allele and can be used for the detection of the wheat technological quality.
