Dominance, selection and cooperation between ovarian follicles in vitro

The aim of pressent work was to develop an in vitro model of co-culture of porcine and bovinne ovarian follicles,to analyze the interrelationships betwen ovarian follicles, their growth and influence of insulin-like growth factor I (IGF-I), oxytocin (OT) and antisera against IGF-I and OT. We have demonstrated the growth of ovarian follicles in vitro, the existence of follicular dominance (inhibition of growth of neighbouring follicles) by bovinne follicles and cooperation (reciprocal support of growth) betwen porcine follicles.Addition of IGF-I or OT mimicked the effect of co-culture stimulation of follicular growth whilst this effect of co-culture was prevented by antisera against IGF-I and OT. Antibody against OT blocked IGF-I effect, whilst antibody against IGF-I inhibited stimulatory effect of OT. This observation suggest, that IGF-I and OT can be mediators of communication betwen porcine ovarian follicles and that OT can stimulate follicular growth throught local IGF-I production, whilst IGF-I can do it throught activation of OT release
