Transient transformation assays for the analysis of gene expression are used as alternatives to genetic complementation and stable plant transformation. Although such assays are routinely performed in several model plant species, they have been rarely applied to flax. Here we tested GFP-based reporter system on different types of flax tissues and organs, such as hypocotyls, cotyledons and leaves, by using two independent transient transformation techniques. When flax leaves were agroinfiltrated with GFP-construct targeted to the endoplasmic reticulum (ER), transiently transformed cells were obtained and ER was visualized by confocal laser scanning microscope analysis. Further, transient expression of cytoskeleton reporter constructs, such as GFP-ABD2 and GFP-MBD was followed in individual epidermal cells of flax cotyledons and hypocotyls transformed by particle bombardment method.
