Efficacy of diazepam in birds: Dose-response study

Effect of diazepam was investigated in 120 budgerigar following intramuscular administration. Six groups of 20 budgerigar differently received 0.10, 0.15, 0.20, 0.25, 0.30, and 0.40 mg. of diazepam per 30 g. body wt. Within 1 to 4 minutes postadministration, 18-20 (90-100 %) birds of each group were sedated and ataxic which lasted 47 to 91 minutes. Hypnosis as indicated by prostration was observed in 8, 6, 11, 11, 12, and 17 birds receiving 0.10, 0.15, 0.20, 0.25, 0.30, and 0.40 gm./30 g. body wt. respectively. Increase in dose rate from 0.10 to 0.25 mg./30 g. body wt. increased duration of hypnosis from 28 to 48 minutes while onset was shortened from 10 to 5.5 minutes. Higher dose rates (0.30 and and 0.40 gm./30 g. body wt.) could not increase the hypnotic duration longer than 48 minutes which was conclusively the efficacy of diazepam in bud gerigar. Hypnotic ED 50 of diazepam in budgerigar was 0.17 mg./30 g. body wt. Diazepam slightly decreased pain response of budgerigar. One bird had recovered from 0.40 gm./30 g. body wt. dose rate and died of starvation 11 hours postadministration. Due to good hypnotic efficacy and safety of diazepam, dose rate of 0.25 mg./30 g. body wt. is recommended for restraining birds subject ot surgery clinically.
