Induction of polyploid from anther-derived plants in tobacco

Induction of polyploid plants from anther-derived plants which have haploid chromosomes was conducted at Mae Jo Tobacco Experiment Station, Chiang Mai, from July 1985 to June 1986 by using the following methods: a) the seedlings were soaked in 0.2% and 0.4% colchicine solution for 4, 18, 24, 48 and 72 hours, after that the seedlings were planted in the pots. b) After transplanting; dropping 0.2% and 0.4% colchicine solution on the seedlings four times a day for two weeks. c) Attach the cotton ball saturated with 0.4% colchicine solution to the axillary buds and flowering buds on the plants at the flowering stage for 24, 48 and 72 hours. 6.66% and 3.33 of diploid plants were induced by soaking the seedlings in 0.4% colchicine solution for 24 and 48 hours, respectively. Dropping 0.4% colchicine for two weeks induced 6% diploid plants. The lethal effects of seedlings by soaking in 0.2% and 0.4% colchicine solution for 72 hours were 13.33% and 33.33%, respectively.
