Genome classification of bananas by chemotaxonomy

Isozyme extracted from 107 bananas leaf samples in phosphate buffer 0.05 M pH 7.5 were separated into an attempt identify Musa acuminata Colla, M. balbisiana Colla, M. ornata Roxb., M. laterita Roxb., M. itinerans Chesseman and M. acuminata X M. balbisiana hybrids. Polyacrylamide vertical slab gel electrophoresis at 7.5 percent concentration was used for peroxidase separation and at 10 percent concentration was used for glutamic oxaloacetic transaminase separation. The peroxidase band of PRX-1 locus and PRX-6 locus could distinguish the banana of AA AAA and AAB genomes from the ABB ABBB and BBB genomes. Also they could distinguish the ABB genome from the ABBB and the BBB genomes. The band of glutamic oxaloacetic transaminase at GOT-2 locus could distinguish the banana of AA AAA and AAB genomes from the ABB ABBB BB and BBB genomes and they could distinguish the ABB ABBB BB genomes from the BBB genomes. The identification of different banana have confronted the complexity of the zymogram patterns of the isozyme, probably due to the difference in developmental stages. Moreover, the resolution of isozyme from two millimeter thick gel appeared to be insufficient for separation of the close bands
