Effects of stevioside and steviol on human chromosomes

Effects of stevioside and steviol on chromosomes from peripheral blood lymphocytes were investigated. Blood samples obtained from 5 healthy donors were cultivated for 72 hours. The medium contained RPMI 1640 3 ml 20 % fetal calf serum and 0.05 ml phytohemagglutinin. After 24 hours of incubation, stevioside 1, 5 and 10 mg/ml or steviol 0.1 and 0.2 mg/ml were added into experimental cultures. Then they were incubated for another 48 hours. After chromosomal preparation, the chromosomes were stained with ordinary Giemsa staining technique and 100 good quality metaphases were analysed. Structural aberrations of the chromosomes were analysed under microscope and recorded in the protocol. The number of cells with aberrations from 100 cells and percent of aberrations from all analysed chromosomes were summarized and compared with control culture. The results from statistical evaluation indicated that stevioside at concentration of 1 mg/ml showed no difference in the frequency of chromosomes aberrations between control and more experiment should be performed. In the cultures contained steviol 0.1 mg/ml expressed no difference in the frequency of chromosome aberrations between control and experimental cultures. Steviol at concentration of 0.2 mg/ml also showed no difference, except from one donor expressed no aberration from all analysed chromosomes. The activation of steviol with S-9 mix was performed only one experiment and one donor. So the sample was too small, anyhow the difference in the frequency of chromosome aberrations between control and tested cultures was not significance.
