Electric fusion of zona-free mouse two-cell embryos

Present study was undertaken to find a suitable condition for blastomere fusion of mouse two-cell embryos using electric fusion to simplify the nuclear transfer procedure. ICR and F1 (C57BL/6J x CBA/N) strain mouse zona-free two-cell embryo blastomeres were paired and treated with electric stimulus under different conditions. Five hours after electric treatment, fused blastomere pairs were embedded in alginate gel and cultured for 96 hr to observe their developmental potential. There was no difference in the fusion rate after stimulus of 30 to 90 micro sec at 1.0 to 2.0 kV/cm and 60 to 90 micro sec at 0.5 kV/cm field strength (77.1-100 %). However, when 30 micro sec at 0.5 kV/cm was used, fusion rate was low (57.1 %) in comparison with the preceding conditions. Blastocyst developmental rate after in vitro culture of the fused blastomer pairs was not different from all the above conditions (81.8-100 %). Chromosome count from well-spread metaphase plates indicated uniform tetraploid.
