Cloning of xylanase gene from Streptomyces griseo-ruber 42-9 into glucose isomerase-producing Streptomyces cyaneus 190-1

Strain improvement of Streptomyces cyaneus 190-1, a glucose isomerase-producing strain, to have ablility to produce xylanase was performed. Xylanase gene from Streptomyces griseo-ruber 42-9 was cloned into Streptomyces cyaneus 190-1 via Streptomyces lividans TK 64 as an intermediate host using pIJ 699 as a cloning vector. Three positive clones giving recombinant plasmids, namely pPT 6 C, pPT 6 E and pPT 6 G with the insert sizes of 4.8, 14.2 and 9.4 kb, respectively, were obtained. Streptomyces cyaneus 190-1 harboring these plasmids were able to produce xylanase at a level comparable to that of Streptomyces griseo-ruber 42-9.
