Lactose hydrolysis in milk using immobilized Beta-Galactosidase

Immobilization of Beta-galactosidase on sand with partical size 35-50 mesh was prepared. Sand was activated by reflux with 3 percent by volume of APTS in toluene for 24 hr. The resulted sand-APTS was further treated consecutively with 1 percent by volume of glutaraldehyde at ph 9 (the intermolecular cross-linker) and 3 percent by volume of beta-galactosidase solution at pH 6.5. The optimum temperature and pH of immobilized enzyme were 20-25 deg C and pH 7-7.5 while 30 deg C and pH 7.5-8 were optimum for the free enzyme. The degree of lactose hydrolysis in milk using the immobilized beta-galactosidase (25 g/100 ml of milk) at room temperature for 16 hr and at 8+-2 deg C for 50 hr were 95 and 99 percent, respectively.
