Effect of MS macro salt and sucrose with two levels of NAA on micropropagation of rejuvenated lilac

Experiment was undertaken to study the combined effect of three factors (MS macro salt, sucrose and NAA) on microporpagation of rejuvenated nodal explants of lilac (Syringa vulgaris L.). Two levels of NAA (0.1 and 1.0 mg/l) were added with all combinations between MS macro salt at 0, 0.5, 1.0 *standard strength and sucrose at 0, 10, 35 g/l respectively. A high concentration of NAA (1.0 mg/l) had a negative influence on rooting while a low concentration of NAA (0.1 mg/l) enhanced the best rooting performance. The negative effect of high auxin was less pronounced in the 0.5/10 and 1.0/35 treatments. The optimal balance was determined as MS macro at half standard strength with sucrose at 20 or 35 g/l and NAA at 0.1 mg/l. However it was assumed that a proper balance between the nutritional factors and regulatory substances is essential for stimulating micropropagation of lilac in vitro.
