Production of cyclodextrins from Bacillus sp. PS304

Bacillus sp. PS304 secreted extracellular cyclodextrin glycosyltransferase (CGTase) to produce cyclodextrins (CDs) from starch. The CDs concentration was quantitated using HPLC since it was a sensitive and reliable method. The optimum conditions for production of CDs were a pH 6.5 at temperature of 50 deg C using 4 percent soluble starch from Merck. The production of alpha, beta and gamma-CDs from Bacillus sp. PS304 in a fermentor was in the ratio 1:4.7:1.2 indicating that beta-CD was the predominant product. The production of CDs was growth associated and reached a maximum at 8 hours of cultivation.
