Comparison of three levels of centrifugation on Thai Ridgeback dog semen cryoconservation

Twenty ejaculates from five Thai Ridgeback dogs were diluted with Egg yolk-Tris diluent at rate of 1:3 to 1:5 depending upon sperm concentration. Each diluted semen was equally divided into 4 treatments; treatment 1: non-centrifugation, treatment 2: centrifuge at 1800 rpm for 10 minutes, treatment 3: centrifuge at 3000 rpm for 10 minutes and treatment 4: centrifuge at 4500 rpm for 5 minutes. After centrifugation, the supernatant was removed. Each pellet was resuspened in Egg yolk-Tris diluent at the same ratio. All the treatments were performed in deep frozen semen. After 24 hours stored in the liquid nitrogen, the post-thaw sperm quality were determined. It was found that the post-thaw motility and the post-thaw percentage of dead spermatozoa of the all centrifuged groups were significantly higher than non-centrifuged group but there were no significant difference among the three levels of centrifugation. The post-thaw normal acrosome of the centrifuged semen at 3000 rpm for 10 minutes and 4500 rpm for 5 minutes worse than that of the centrifuged semen at 1800 rpm for 10 minutes and that of non-centrifuged semen.
