Responding of the Lemmi 9 promoter in transgenic potatoes to Meloidogyne incognita infection

Transgenic potatoes carrying a T-DNA with P Lemmi9-gus were grown in vitro and were inoculated with Meloidogyne incognita. GUS activity can be detected immediately one day after inoculation. By embedding and sectioning the GUS stained tissues, the results indicated that the Lemmi9 promoter is not only induced upon giant cell formation but also in the cells nearby the nematode feeding sites (NFSs). Extracted DNA from 6 lines of transgenic potatoes were digested with either Nsil, Scal or both of the restriction enzymes. The DNA fragments were electrophoresed in 0.8 percent agarose gel and transferred to nylon membrane by Southern blotting. DNA fragments containing gus were visualized by hybridizing with a 32P-labelled GUS probe. The copy numbers of T-DNA were found to be different among the lines. These results implied that the more copy numbers appeared the more degree of expression revealed.
