This project aims that find out suitable viability test in order to determine right seed collection time, most appropriate method for protokorm formation and possibilities of using artificial seed by alginate matrixes on the production. It is found that Fluorescein diacetate (FDA) application on the isolated embryo did not give off sufficient florescent light under UV to judge viability rate. Knodson C, is growth media, was found to formed protocorm and Alginate-Chitosan can be used safely to set artificial seed.
Show more [+] Less [-]Turkish. Salep olarak kullanılan Orchis italica,ve Serapias vomeracea türlerinde uygun canlılık testi yöntemini saptayıp, en uygun tohum alma zamanını belirlemek. İn vitro teknikler kullanarak en uygun protokorm oluşturma yöntemlerini belirlemek. Protokormların alginate matriksleri ile kaplayarak suni tohum elde etmek ve bunların üretimde kullanılabilme olanaklarını ortaya koymak amacıyla yürütülen bu çalışmada; Floresan diasetat (FDA) ile muamele edilen embriyolardan elde edilen flüoresans ışıma bu türlere ait tohumlarda çanlılık testi için yeterli olmadığı saptanmıştır. Protokorm oluşturmada Knodson C sıvı ortamının kullanılabileceğini ve oluşturulan protokormların Alginate-Chitosan ile kaplanarak suni tohum oluşturula bileceği bulunmuştur.
