Determination of vegetative compatibility groups (VCGs) of Verticillium dahliae kleb. isolates obtained from cotton growing areas of aydin province | Aydın ilinde pamuk alanlarından elde edilen Verticillium dahliae kleb. izolatlarının vejetatif uyum gruplarının (VCGs) saptanması

English. The study was carried to determine the Vegetative Compatibility Groups (VCGs) of Verticillium dahliae isolates obtained from the cotton growing areas of Aydın province. In survey studies carried out 12 important cotton-growing counties of Aydın in 2004-2005, 47 Verticillium spp. isolates were obtained and identified as Verticillium dahliae Kleb. Pathogenicity tests were performed on cotton plants cv. Acala SJ2. The virulence of the isolates changed between 3.83 and 100% while the percentage of leaf shedding ranged from 0 to 100. 64 mutants obtained from 24 single spore V.dahliae isolates were tested with tester strains for heterokaryosis. While 38 out of 54 mutants obtained from the cotton fields were VCG2B, 6 was strongly compatible with VCG2B but weakly compatible with VCG1, 5 of them was strongly compatible with VCG2B and VCG1, 2 was strongly compatible with VCG2A, 1 was strongly compatible with VCG1 and weakly compatible with VCG2B, 1 was weakly compatible with VCG2B and VCG4B, and 1 was weakly compatible with VCG2B.

Çalışma, Aydın İli pamuk ekim alanlarından elde edilen Verticillium dahliae izolatlarının Vegetatif Uyum Gruplarını (VCGs) tespit etmek amacıyla yapılmıştır. 2004-2005 yıllarında Aydın ilinde pamuk ekim alanı açısından önemli 12 ilçede yapılan survey çalışmalarında, 47 Verticillium spp. izolatı elde edilmiş ve hepsi Verticillium dahliae Kleb.olarak tanılanmıştır. Acala SJ2 pamuk çeşiti ile yapılan patojenisite çalışmalarında izolatların virülensi %3.83-100 arasında değişmiştir. İzolatların pamuk bitkilerindeki yaprak döküm oranlarının ise %0.0-100.0 arasında değiştiği belirlenmiştir. 24 V.dahliae tek spor izolatından elde edilen 64 mutant tester izolatları ile heterokaryosis testlerine alınmıştır. Pamuk alanlarından elde edilen 54 mutantın 38'i VCG2B iken, 6'sı VCG2B ile kuvvetli fakat VCG1 ile zayıf, 5' i VCG 2B ve VCG1 ile kuvvetli, 2'si VCG2A ile kuvvetli, 1'i VCG1 ile kuvvetli fakat VCG2B ile zayıf, 1'i VCG2B ve VCG4B ile zayıf, 1'i ise VCG2B ile zayıf heterokaryosis oluşturmuştur.
