Investigation of effects of chaste tree (Vitex agnus-castus L.)on diabetic wound healing in rats and antibacterial properties

In this thesis study, the effect of plant extract on diabetic wound healing of Vitex agnus-castus L. (Hayıt), which has an important medical significance, and allelopotential effects of secondary metabolites of this plant were searched. GCMS which left total phenolic contents and Gr (-) Escherichia coli ATCC 25922, Gr (-) Pseudomonas aeruginosa ATCC 27853, Gr (+) Staphylococcus aureus ATCC 25923 and Gr (+) Micrococcus luteus NCIMB 13267 bacteriums' antimicrobial effects were identified by disk diffusion method. In the wound healing experiment, 30 male and 30 female totally 60 healthy rats, which are Wistar-Albino genera, were used. Their average weights were 230 g (200-250 gr) for 20 weeks. Male and female rats were divided into five groups of 65, 265, 465, sham and control groups has six rats in each group. The extracts prepared for 21 days were applied to the wound area by injection into the rats. As a result, differences were observed in the wound healing process of male and female rats, and as the amount of dose increased, the rate of wound healing in males increased. In females, however, there was a rapid improvement compared to the differenet doses, the best healing was 265 mg. Finally, antimicrobial effects of leaf and seed oil on both Gr (+) and Gr (-) bacteria was observed.
