The Czech barley variety Diamant has been created from the variety Valticky by X-ray radiation. RAPD, SSR and AFLP methods were used to find out differences on the DNA level between these two varieties. The genetic polymorphism between the two varieties was as follows: 39% (RAPD), 42.8% (SSR), 11.4% (AFLP). These results indicate either big mutations caused by the irradiation or a high genetic heterogeneity of the original variety. A considerable discrepancy was found between molecular and pedigree data, thus confirming the importance of molecular methods. The variety Diamant showed higher levels of extract content and proteolysis than the variety Valticky. On the contrary, the variety Valticky exhibited higher values of thousand-grain weight and volume weights of grain and malt.
