Human Serine Hydroxymethyltransferase 2 (SHMT2) linteracts with hnRNP D

Heterogeneous nuclear ribonucleoprotein D (hnRNP D) is one of the principal pre-mRNA binding proteins present in human cells. It is a ubiquitously expressed, nuclear protein. The protein contains two RNP-consensus RNA-binding domains and a glycine-rich C-terminal domain. It has been known to be associated with the regulation of mRNA stability by binding to AU-rich elements (ARE) present in the 3' UTR of several mRNAs. In order to identify cellular proteins that bind to hnRNP D, this research conducted yeast two-hybrid screening using a HeLa cDNA library. It then discovered that one of the cDNA clones carried a partial human serine hydroxymethyltransferase 2 (mitochondrial) (SHMT2) cDNA (GenBank accession number NM_005412). This paper shows for the first time that human serine hydroxymethyltransferase 2 binds specifically to hnRNP D, both in yeast two-hybrid system and in vitro pull-down assay.
