Transfering of virus free plantlets from invitro to invivo condition.

This research was carried out with the purpose of investigating the effect of different planting beds and the position of stem nodes on the production of minituber from tissue culture plantlets in two varieties of potatoes. For this purpose,the tubers from &ldquoMarfona&rdquoand &ldquoAgria&rdquowere planted in the greenhouse. When the plants were 15-20 cm, they were toped and the plants were transfered to thermotherapy conditions. After four weeks, the meristems were excised and placed on paper bridge in liquid media and the resulted plantlets were tested for virus free condition. In order to propagate the virus free plantlets, three liquid culture media were investigated. They were : 1-MS(without hormones) 2-MS+0.5mg/lit BAP 3-MS+0.5mg/L BAP+0.05mg/L NAA . Result showed that the media without hormone was the best and produced vigorous plantlets with excellent rooting. Therefor, all plantlets were propagated in hormone free media. Thereafter, in order to investigate the effect of planting beds, the position of stem nodes on minituber production, first, the apical meristem of stems and whole roots were removed and then each stem was divided into three sections (Bottom,Middle and Top) and these sections were cultured on four planting bed:1-peat moss/perlite(3:1) 2-turf/perlite (3:1) 3-leaf mould/perlite (3:1) 4-rice hull/turf (1:1). The results showed that all planting beds except leaf mould/perlite(3:1)produced desirable plants. The position of nodes only had effect on the ratio of compound leaf to total leaves and this was significant. After harvesting minitubers and measuring the traits, it was clear that rice hull/turf (1:1) planting bed, in all traits was better than other planting beds. In this stage, effect of the position of nodes except for the interaction between position of nodes×planting bed on total minituber weight was not significant.
